From: Synthetic RNA devices to expedite the evolution of metabolite-producing microbes

RNA aptamer pools specific for target molecules are constructed by systematic evolution of ligand by exponential enrichment (SELEX). Using these in vitro-selected aptamers, a Riboselector is generated in vivo through several rounds of dual positive and negative selection. After determining the selection condition for the parental strain using the Riboselector, the generated library can be enriched under the chosen selection pressure. Iterative strain improvement can be achieved by re-setting the selection conditions or re-constructing a Riboselector suitable for the newly developed parental strain.
