HDAC6 modulates acetylation of SP1 as well as its binding to the EWSR1/EWSR1-FLI1 activating promoter regions. In turn, HDAC6 increases SP1/p300 complex binding by deacetylating SP1. HDAC6 inhibition allows dissociation of the SP1/p300 complex by increased acetylation of SP1, with a subsequent decrease in both the expression of EWS fusion gene transcription and its protein activity, leading to modulation of a specific EWS target genes.
