1 × 105 naïve TCR7B8 CD4 T cells collected from spleen and LNs of TCR7B8 mice, TCR1A2 Rag2−/− mice, or TCROTII TCRα−/− mice, were retro-orbitally transferred into SFB-colonized Rag2−/− hosts. TCROTII transferred Rag2−/− hosts were maintained with 4 mg/ml Ovalbumin containing water. Frequency of natural Tregs (Gated as DAPI-, TCRβ+, CD8α-, CD4+, Foxp3+, and RORγt-) (A) and inducible Treg (Gated as DAPI-, TCRβ+, CD8α-, CD4+, Foxp3+, and RORγt+) (B). Numbers of Foxp3+ IL-10 producing CD4 T cells (C), Foxp3- IL-10 producing CD4 T cells (D), and IFNγ and IL-10 double producing CD4 T cells (Tr1 cells) (E). Two independent experiments were combined. Results are presented as the mean ± SEM, with dots representing individual mice. Two-way ANOVA. (C, D, E) during cell preparation, one brain sample of 1A2 transferred and one spleen samples of OTII transferred mice were lost.
