Effect of pH, temperature, organic solvents and detergents on activity of PsChO. (A) pH dependence of PsChO (Left): Activity was measured in the reaction mixtures adjusted to various pHs with 25 mM MES, PBS, HEPES, Tris-HCl, and glycine-NaOH buffers, respectively. Analysis of pH stability of PsChO (Right): The PsChO was pre-incubated at different pH values for 1 h at 4 °C, and the residual activity was determined in the standard assay conditions. (B) Temperature dependence of PsChO (Left): Activity was measured at various temperatures in the standard assay conditions. Thermostability analysis of PsChO (Right): The PsChO was pre-incubated at various temperatures for 30 min, and the residual activity was determined in the standard assay condition. (C) Organic solvents and detergents were added to the solution at 4 °C for 1 h. The relative residual activity shows the activity as compared to that observed in a control PsChO solution not exposed to the organic solvent/detergents.
