IR and LDLR knock down by siRNA attenuates autophagy inhibition by LDL. HUVECs were transfected with IR-siRNA, LDLR-siRNA or scrambled siRNA for 48 h then starved by culture in serum-free medium overnight. After successful silencing, cells were treated with or without LDL (50 μg/mL) for 60 min then cell lysates analyzed by Western blotting. The expression of IR, LDLR, autophagy-associated proteins, and PI3K/Akt/mTOR and PI3K/Akt/GSK3β pathway proteins were analyzed. The expression of Ctr group cells was assigned the value of 1, n = 3, *p < 0.05, **p < 0.01 versus scrambled siRNA alone. #p < 0.05, ##p < 0.01 versus scrambled siRNA + LDL. Data expressed as mean ± S.E.M.
