Monitoring the peptide-toxin complex formation via gel-filtration. (A) Gel-filtration profile of αBgt (green), HAP (orange) and their mixture (blue) on the Superdex 75 10/300 GL column. Pure HAP and αBgt are well separated on this column, thus allowing to monitor the formation of the complex between HAP and three-finger toxin (see Fig. S10 for the LC-MS). (B) No signs of HAP-αCtx complex formation observed on the gel-filtration profile of their mixture. (C) HAP[L9E] analogue of HAP interacts with αBgt in a concentration-dependent manner as can be monitored via reduction of the respective peak on gel-filtration profile with increasing concentrations of αBgt. (D) HAP[L9E] analogue of HAP also interacts with αCtx in a concentration-dependent manner and reduction of its peak can be monitored on the gel-filtration profile with increasing concentrations of αCtx.
