In the context of HFD-induced obesity, osteoblastic 11β-HSD1 levels are upregulated, leading to overactivated glucocorticoid (GC) signaling. This augmented GC signaling downregulates the expression of Early Growth Response 2 (Egr2) in osteoblasts, and the suppressed Egr2 further restrains skeletal glucose uptake and bone formation. Consequently, the reduction in skeletal glucose uptake and bone formation contributes to bone loss and glucose handling impairment associated with obesity. This figure was created in BioRender.
