Testing the ___location of regions of homology. (A) Both regions have exactly 50 base pairs of homology, however; in the top scheme this is included entirely on the insert allowing the same vector PCR product to be used for any insert. In the bottom scheme, the homology is divided between the insert and the vector, with the exact split determined by the melting temperature of the amplification site, preventing parallelization. The base pairs for primers to construct pPICZαA[PLAT] have been included as an example.
