Optimization of Au and HA concentration in Au-PEG-HA NPs for cell detection. (a) Colorimetric changes (with A650/A521 ratio) of Au-PEG-HA NPs after incubation with MDA-MB-231 (50,000 cells) and BT-474 (50,000 cells) at varied Au concentration (100–3.125 nM) show pink-violet shift (521 nm–559 nm) in MDA-MB-231 and no shift in BT-474. (b) UV/Vis absorbance spectrum analysis of Au-PEG-HA NPs after incubation with MDA-MB-231 (50,000 cells) and BT-474 (50,000 cells) at 25 nM Au concentration. (c) Colorimetric changes (with A650/A521 ratio) of Au-PEG-HA NPs after incubation with MDA-MB-231 (50,000 cells) and BT-474 (50,000 cells) at varied HA concentration (400–1.56 µg/ml) and 25 nM Au. (d) UV/Vis absorbance spectrum analysis of Au-PEG-HA NPs after incubation with MDA-MB-231 (50,000 cells) and BT-474 (50,000 cells) at 100 HA µg/ml concentration (25 nM Au).
