MIF enhanced the cytotoxic effects of MCDC in U251 cells. (A) Relative expression levels of endogenous MIF in MCF7 and U251 cells. The expression level of MIF was measured by qRT-PCR. Internal control: ACTB. The relative expression level of MCF7 cells was normalized to that of U251 cells. (B) Overexpression of MIF in U251 cells. MIF was overexpressed and measured by qRT-PCR 24 h after transfection with the amounts of plasmid shown. (C) Effects of MIF on drug sensitivity of MCDC in U251 cells. Different amounts of MIF were overexpressed in U251 cells treated with different doses of MCDC. The viability of cells was measured by MTT assays. (D) Effects of MIF N6Q/T7M mutant on viability of U251 cells treated with MCDC. The viability of U251 cells overexpressing MIF or MIF mutants under different doses of MCDC was measured by MTT assays.
