Determination of cell-to-cell spread by plaque assays. DEF cells in 6-well plates were infected with 0.001 MOI of BAC-CHv, BAC-CHv-ΔUS3 or BAC-CHv-ΔUS3R. After incubation at 37 °C for 2 h, the infected cells were covered with 1% methylcellulose and cultured. (a) Green fluorescent plaques produced by the US3-deleted, parental and revertant viruses. (b) and (c) Statistical analysis of twenty different randomly selected viral green fluorescent plaques. (d) Images of viral plaques after 1.5% crystal violet staining.
