General scheme of the immunoassay. IsdB is immobilized on the bottom of a StrepTactin®XT 96-well plate exploiting its C-terminal StrepTag®II. Hb, alone (top scheme) or after pre-incubation with the compounds at 1 mM concentration (bottom scheme), is then added to the wells. After washing steps, the antiHb HRP-conjugated antibody is added, followed by the substrate 3,3′,5,5′-tetramethylbenzidine (TMB). The reaction is blocked by H2SO4 and the signal intensity is measured at 450 nm.
