KLF4 is involved in the organization and regulation of pluripotency-associated three-dimensional enhancer networks

3D Enhancer–promoter networks provide predictive features for gene expression and coregulation in early embryonic lineages

Here, the authors describe 3D hubs as regulatory subunits of gene expression in the three essential lineages of embryogenesis. They develop a computational model that can predict novel enhancers and they validate such enhancers in the context of specific lineages.

A metabolic interplay coordinated by HLX regulates myeloid differentiation and AML through partly overlapping pathways

HLX transcription factor regulates haematopoietic stem and progenitor cell (HSPC) differentiation and is overexpressed in acute myeloid leukemia. Here the authors show that HLX overexpression leads to myeloid differentiation block in zebrafish and human HSPCs by direct regulation of metabolic pathways.

Chronic p53-independent p21 expression causes genomic instability by deregulating replication licensing

Galanos and colleagues observe p21 accumulation in proliferating cancer cells, and show that in cultured p53-null cells, p21 can cause genomic instability by perturbing replication licensing through inhibition of the CRL4-CDT ubiquitin ligase.
