The chromatin remodeling enzyme Chd4 regulates genome architecture in the mouse brain

The mechanisms underlying gene regulation and genome architecture remain poorly understood. Here, the authors investigate the role of chromatin remodelling enzyme Chd4 in granule neurons of the mouse cerebellum and find that conditional knockout of Chd4 preferentially activates enhancers and modulates genome architecture at a genome-wide level.

Astrocyte deletion of α2-Na/K ATPase triggers episodic motor paralysis in mice via a metabolic pathway

Mutations of α2-Na/K ATPase can cause familial hemiplegic migraine via unclear mechanisms. Here, the authors show that deletion of α2-Na/K ATPase in astrocytes results in gene expression and metabolic changes leading to cortical spreading depression and episodic transient motor paralysis in mice.

A CaMKIIβ signaling pathway at the centrosome regulates dendrite patterning in the brain

This study reports that CaMKIIβ is recruited to the centrosome by PCM1, where it promotes dendrite retraction and pruning via the phosphorylation and inhibition of Cdc20-APC. This effect is independent of its association with CaMKIIα.

CHARGE syndrome protein CHD7 regulates epigenomic activation of enhancers in granule cell precursors and gyrification of the cerebellum

CHARGE syndrome that affects cerebellar development can be caused by haploinsufficiency of the chromatin remodeling enzyme CHD7; however the precise role of CHD7 remains unknown. Here the authors show CHD7 promotes chromatin accessibility and enhancer activity in granule cell precursors and regulates morphogenesis of the cerebellar cortex, where loss of CHD7 triggers cerebellar polymicrogyria.

An α2-Na/K ATPase/α-adducin complex in astrocytes triggers non–cell autonomous neurodegeneration

It has been suggested that astrocytes play a role in the onset and progression of neurodegenerative disorders such as ALS. In this study, the authors show that α-adducin forms a complex with α2-Na+/K+ ATPase in mutant SOD1-bearing astrocytes and that this interaction is necessary for the non-cell autonomous toxicity that induces muscle denervation, motor neuron death and decreased mortality.

Control of glioblastoma tumorigenesis by feed-forward cytokine signaling

Glioblastoma is a deadly brain tumor with no cure. The cytokine receptor OSMR is identified as a new key player in glioblastoma pathogenesis. OSMR orchestrates a feed-forward mechanism with the oncogenic protein EGFRvIII and the transcription factor STAT3 to drive oncogenesis. Loss of OSMR impairs EGFRvIII-STAT3 signaling and glioblastoma tumorigenesis.

A molecular basis for phosphorylation-dependent SUMO conjugation by the E2 UBC9

Phosphorylation-dependent SUMOylation of MEF2A promotes postsynaptic dendrite differentiation. Analyses now reveal that a surface on the SUMO E2 UBC9 is responsible for integrating phosphorylation signal recognition and SUMOylation and suggests that regulation of some SUMO substrate recognition events may have evolved to use the E2 rather than an E3 ligase.
