LNP-RNA-mediated antigen presentation leverages SARS-CoV-2-specific immunity for cancer treatment

Immunoengineering-based cancer therapies have huge potential. Here the authors report on the lipid nanoparticle delivery, to cancer cells, of self-amplifying RNA encoding SARS-CoV-2 spike epitope-loaded MHC I molecules to take advantage of anti-SARS-CoV-2 immunity from a vaccinated population to treat cancer.

Difunctionalization of bicyclo[1.1.0]butanes enabled by merging C−C cleavage and ruthenium-catalysed remote C−H activation

The high fraction of sp³-hybridized carbon atoms (Fsp³) in cyclobutanes makes them promising bioisosteres for arenes. Now by combining bicyclo[1.1.0]butane C–C activation with remote meta-C–H functionalization, via a ruthenacycle-mediated halogen-atom transfer process, densely decorated Fsp³-rich cyclobutanes can be formed in a single step with high chemo- and position selectivity.

SEPTIN2 suppresses an IFN-γ-independent, proinflammatory macrophage activation pathway

Interferon-gamma (IFN-γ) is an important but not exclusive proinflammatory mediator in macrophages. Here authors show that IFN-γ-independent macrophage autoactivation involves endoplasmic reticulum (ER) stress which in turn induces the GTP-binding protein Septin2 to limit inflammation via a negative feedback loop.
