Hematopoietic stem and progenitor cell-restricted Cdx2 expression induces transformation to myelodysplasia and acute leukemia

The caudal-related homeobox transcription factor CDX2 is expressed in leukemic cells in the majority of patients with leukemia but not during normal blood formation. Here, the authors report a mouse model with conditional Cdx2 expression showing de novo leukemic transformation, and use it to optimize treatment in high-risk AML.

TOPORS E3 ligase mediates resistance to hypomethylating agent cytotoxicity in acute myeloid leukemia cells

Hypomethylating agents (HMAs) are frontline therapy for Myelodysplasia, but their efficacy is limited. Here, the authors show that depleting the dual E3-ligase TOPORS synergizes with HMAs by enhancing DNA damage in leukemia cells, improving survival in mouse models without preventing healthy hematopoiesis.

Prostaglandin E2 regulates vertebrate haematopoietic stem cell homeostasis

This study identifies Prostaglandin E2 (PGE2) as an important modulator of vertebrate haematopoietic stem cell (HSC) number. Chemicals that enhance PGE2 synthesis increase the number of HSCs whereas the opposite effect is achieved by blocking PGE2 synthesis.

Marshall et al. show that regions of the genome adopt a noncanonical Z-DNA state in the prefrontal cortex in response to fear learning and that binding of Adar1 reduces Z-DNA during extinction learning, which is required for memory flexibility.

ADAR1 is an adenosine deaminase that acts on double-stranded RNA. Orkin and colleagues show that ADAR1 protects hematopoietic stem cells from interferon-induced insult and is needed to maintain long-term hematopoiesis.

Developmental and species-divergent globin switching are driven by BCL11A

The contribution of changes in cis-regulatory elements or trans-acting factors to differences in gene expression between species is not well understood. Here it is found that, in transgenic mice containing the human β-globin locus, the expression of BCL11A differs between mouse and human and is a critical mediator of species-divergent globin switching.
