Involvement of a chromatin remodeling complex in damage tolerance during DNA replication

Remodeling complexes can affect DNA transactions by altering chromatin, thus affecting accessibility of DNA. The INO80 remodeling complex has previously been implicated in replication and analyses now argue that it specifically acts through the DNA damage tolerance pathways that resolve recombination intermediates at impeded replication forks.

The histone chaperone SPT2 regulates chromatin structure and function in Metazoa

Using a combination of bioinformatics, biochemistry, genetics, genomics and cell-based approaches, this study shows that the H3–H4 binding capacity of the histone chaperone SPT2 is required to preserve chromatin structure and function in Metazoa.

Supervised and unsupervised deep learning-based approaches for studying DNA replication spatiotemporal dynamics

When tested on well-characterised cellular models, supervised and unsupervised deep learning closely recapitulate expected differences in DNA replication dynamics across genotypes, holding promise for identifying aberrant DNA replication at scale.

Chromatin replication and epigenome maintenance

The influence of chromatin structure on the DNA replication programme is reciprocated by replication-coupled mechanisms that re-establish chromatin on newly formed DNA. The tight coupling of these processes is essential for promoting integrity of the genome and epigenome, with possible implications for ageing and cancer.

H4K20me0 marks post-replicative chromatin and recruits the TONSL–MMS22L DNA repair complex

We have a limited understanding of how cells mark and identify newly replicated genomic loci that have a sister chromatid; here, unmethylated K20 in the tail of new histone H4 is shown to serve as a signature of post-replicative chromatin, which is specifically recognized by the homologous recombination complex TONSL–MMS22L.

Groth and colleagues identify proteins associated with newly synthesized DNA — isolated by nascent chromatin capture — as well as proteins associated with mature DNA, and find factors not previously linked to replication or nascent chromatin.
