6mA-DNA-binding factor Jumu controls maternal-to-zygotic transition upstream of Zelda

N6-methyladenine (6mA) DNA modification is a dynamic epigenetic mark in Drosophila embryos, but how 6mA is decoded is unclear. Here, the authors show that the protein Jumu binds 6mA-marked DNA to regulate the maternal-to-zygotic transition, partly through regulation the expression of the 6mA marked pioneer factor zelda.

PCBP2 maintains antiviral signaling homeostasis by regulating cGAS enzymatic activity via antagonizing its condensation

cGAS senses viral DNA and forms cytosolic cGAS-DNA granules to mediate anti-DNA viral signaling pathway. Here the authors show that PCBP2 interacts with cGAS and antagonizes condensation of cGAS-DNA granules, thus maintaining host immune homeostasis.

Syndecan-4 negatively regulates antiviral signalling by mediating RIG-I deubiquitination via CYLD

Syndecans are transmembrane proteoglycans implicated in diverse cellular activities. Here the authors show that Syndecan-4 via its cytosolic ___domain negatively regulates antiviral immunity by enhancing RIG-I interaction with a deubiquitinating enzyme CYLD, thus inhibiting the activating K63-linked RIG-I ubiquitination.

Arginine methylation of METTL14 promotes RNA N⁶-methyladenosine modification and endoderm differentiation of mouse embryonic stem cells

The methyltransferase complex of METTL3-METTL14-WTAP is responsible for m⁶A modification on RNA. Here the authors report that METTL14 arginine 255 (R255) is methylated by PRMT1 and this modification increases interaction of METTL3/METTL14 interaction with WTAP and substrate RNA, promoting m⁶A methylation activity of the complex.

TERT assists GDF11 to rejuvenate senescent VEGFR2⁺/CD133⁺ cells in elderly patients with myocardial infarction

This study shows how telomerase reverse transcriptase (TERT) assists growth differentiation factor 11 (GDF11) to rejuvenate senescent endothelial progenitor cells via the Smad2/3 and eNOS pathways. Therapeutically, it may be possible to exploit TERT mediated-GDF11 signaling to activate senescent stem cells in the myocardial ischemic microenvironment.
