Activation of the transcription factor NFAT5 in the tumor microenvironment enforces CD8⁺ T cell exhaustion

Verdeil and colleagues show that the transcription factor NFAT5 is selectively required in tumor-induced, but not chronic infection-induced, CD8⁺ T cell exhaustion, possibly due to the modulation of NFAT5 activation by hyperosmolarity in the tumor environment.

Tumour-derived PGD2 and NKp30-B7H6 engagement drives an immunosuppressive ILC2-MDSC axis

Group 2 innate lymphoid cells (ILC2s) modulate inflammatory and allergic responses, but their function in cancer immunity is still unclear. Here the authors show that, in acute promyelocytic leukaemia, tumour-activated ILC2s secrete IL-13 to induce myeloid-derived suppressor cells and support tumour growth.

Regulatory circuits of T cell function in cancer

In this Review, the authors summarize the features and the molecular drivers of T cell dysfunction in cancer and compare these with dysfunctional T cells in chronic viral infection. The metabolic competition in the tumour microenvironment is also discussed. Understanding antitumour T cell responses has important implications for cancer immunotherapy.
