Small circular RNAs as vaccines for cancer immunotherapy

Small circular RNAs encoding one or more antigens and enclosed in subcutaneously injected lipid nanoparticles can elicit potent and durable antitumour T cell responses for robust tumour immunotherapy, particularly in combination with immune checkpoint inhibition, as shown in multiple mouse models of tumours.

Loss-of-function mutations in Keratin 32 gene disrupt skin immune homeostasis in pityriasis rubra pilaris

Pityriasis rubra pilaris (PRP) is a chronic immune‐mediated papulosquamous dermatosis and is caused by dysregulation of keratinocytes. Here the authors identify keratin 32 (KRT32) mutations associated with PRP which results in the inhibitory function of KRT32 towards NF-kB signalling and IKK complex formation being reduced.

Intertwining DNA-RNA nanocapsules loaded with tumor neoantigens as synergistic nanovaccines for cancer immunotherapy

Nucleic acid nanomedicines are promising for cancer drug delivery. Albumin/vaccine nanocomplexes that assemble in vivo for combination cancer immunotherapy

Albumin conjugates can enhance drug delivery. Here, the authors repurpose albumin-binding Evans blue to develop nanovaccines that co-deliver adjuvants and tumor neoantigens to antigen-presenting cells in lymph nodes, resulting in potent and durable antitumour immunity in combination immunotherapy.

Genomic analyses of primitive, wild and cultivated citrus provide insights into asexual reproduction

Qiang Xu and colleagues sequence four citrus species de novo, along with 100 accessions, including primitive, wild and cultivated citrus. Their genomic analyses associate the CitRWP gene with polyembryony and suggest that regions harboring energy- and reproduction-associated genes are probably under selection in cultivated citrus.

Allosteric inhibition of SHP2 phosphatase inhibits cancers driven by receptor tyrosine kinases

Nuclease-resistant synthetic drug-DNA adducts: programmable drug-DNA conjugation for targeted anticancer drug delivery

We report nature-inspired drug-DNA adducts (DDAs) as a simple yet programmable platform for site-specific drug-DNA conjugation and application in targeted anticancer drug delivery. With multiple copies of drugs conjugated on one DNA, the DDAs were nuclease-resistant and stable for storage, yet gradually released drugs at physiological temperature. Designer DDAs were able to form nanoadducts by hybridization and fold into drug-aptamer adducts (DAAs) for specific recognition of cancer cells and targeted anticancer drug delivery. In a tumor xenograft mouse model, DAAs significantly inhibited tumor growth and reduced side effects, as verified by tissue analysis of tumors and hearts.
