Extracellular vesicles in liver disease and potential as biomarkers and therapeutic targets

Extracellular vesicles have emerged as having key roles in liver disease. In this Review, Szabo and Momen-Heravi discuss the functions of extracellular vesicles in liver disease pathogenesis and progression, and explore their potential use as biomarkers, therapeutic targets and tools for therapy delivery.

MicroRNAs in liver disease

This Review summarizes current knowledge on the role of different microRNAs (miRNAs) in liver diseases. The functions of the most relevant miRNAs that have specific roles in liver damage, hepatocyte functions, viral hepatitis, alcoholic and nonalcoholic liver disease, liver fibrosis and hepatocellular carcinoma are highlighted. The emerging utility of miRNAs as potential biomarkers in liver diseases is also discussed.

Women and alcoholic liver disease — warning of a silent danger

Alcohol use and 'social' drinking are increasing among the young and particularly in women. However, gender equity does not extend to the risk of alcohol-associated liver disease and alcoholic hepatitis, which are increased and often caused by as little as half as much cumulative alcohol consumption in women compared with men.

Zhang, Xu, Liu, Wang et al. identify an inhibitory mechanism for RIPK1 kinase through EGLN1/pVHL-mediated proline hydroxylation, which is disrupted upon prolonged hypoxia that activates RIPK1 activity to promote cell death and inflammation.

Steatosis, inflammasome upregulation, and fibrosis are attenuated in miR-155 deficient mice in a high fat-cholesterol-sugar diet-induced model of NASH

Nonalcoholic fatty liver disease is the most common cause of chronic liver disease globally and a risk factor for hepatocellular carcinoma. The study highlights the role of miR-155 in nonalcoholic steatohepatitis. miR-155-deficient mice displayed overall protection from diet-induced steatohepatitis and fibrosis. Therapeutic inhibition of miR-155 might be effective approach for treatment of nonalcoholic steatohepatitis.
