CAD, a de novo pyrimidine synthesis pathway enzyme, is known to promote chemotherapy resistance. Here, the authors shed light into this mechanism of chemoresistance, showing that chemotherapeutic drugs can promote the degradation of CAD following cleavage by activated caspase-3 on its Asp1371 residue.
