Altered extracellular matrix structure and elevated stiffness in a brain organoid model for disease

Brain tissue mechanics influence development and disease. Here, the authors show that LIS1 mutations increase stiffness in human brain organoids due to ECM alterations and that targeted ECM modulation restores mechanical properties.

Correlated structural kinetics and retarded solvent dynamics at the metalloprotease active site

Solvent dynamics are an often overlooked component in enzymatic activity. Terahertz spectroscopy, X-ray absorption analysis and molecular dynamics simulations show that solvent movement is tightly correlated with formation of a productive enzyme–substrate complex, but not an enzyme–inhibitor complex, in a metalloprotease, indicating that solvent motions may assist catalysis.

A multi-modal analysis of pre-metastatic liver biopsies from patients with localized pancreatic cancer with a minimum of 3 years of follow-up shows that immunological, proliferative and metabolomic features distinguish patients who develop metastases from disease-free survivors and can be used to predict outcomes.

Antibodies targeting the catalytic zinc complex of activated matrix metalloproteinases show therapeutic potential

By immunizing mice with a small synthetic metal-baring complex mimicking the active site of matrix metalloproteinases (MMPs), the authors have generated antibodies that bind and inhibit activated MMP2 and MMP9 in a manner analogous to the mechanism of action of tissue inhibitors of metalloproteinases. Targeting the active conformation of these MMPs is therapeutically effective in mouse models of inflammatory bowel disease.

ADAM metalloproteases promote a developmental switch in responsiveness to the axonal repellant Sema3A

The basic axonal wiring of the vertebrate nervous system is set up during embryonic development, and during this process axons gain or lose sensitivity to guidance cues. Here the authors show that in proprioceptive axons, two members of the ADAM metalloproteinase family promote loss of responsiveness to Sema3A, via cleavage of its receptor Neuropilin-1.

Dysfunction of the deacetylase Sirt1 has been associated with certain metabolic diseases. Abramson and colleagues show that Sirt1 has high expression in the thymus, where it deacetylates the transcriptional regulator Aire and is essential for Aire's ability to switch on the expression of tissue-specific genes.
