In the Tumor Profiler proof-of-concept observational study, a multiomics approach for profiling tumors from patients with melanoma was feasible, returning data within 4 weeks and informing treatment recommendations in 75% of cases.

Observation of ultrafast interfacial Meitner-Auger energy transfer in a Van der Waals heterostructure

Here, the authors investigate the interfacial charge/energy transfer dynamics in a WSe₂/graphene heterostructure. They unveil an energy transfer mechanism from WSe₂ to graphene mediated by an interfacial Meitner-Auger process, resulting in a transient hole distribution in the Dirac cone at energies larger than the photon energy of the optical excitation.

Detection of isoforms and genomic alterations by high-throughput full-length single-cell RNA sequencing in ovarian cancer

Long-read single-cell RNA sequencing is capable of detecting isoform-level gene expression and genomic alterations such as mutations and gene fusions, thereby providing cell-specific genotype-phenotype information. Here, the authors use long-read scRNA-seq on metastatic ovarian cancer samples and detect cell-type specific isoforms and gene fusions that may otherwise be misclassified in short-read data.

DNA methylation profiles in monozygotic and dizygotic twins

Arturas Petronis and colleagues present the first genome-wide study of interindividual differences in DNA methylation through genome-scale DNA methylation profiling in tissues from monozygotic and dizygotic twins. The data showed that dizygotic co-twins have higher degrees of epigenetic differences than monozygotic co-twins, which the authors attribute at least partially to epigenetic, as opposed to genetic, differences in the zygotes.

Dynamic gravitational excitation of structural resonances in the hertz regime using two rotating bars

The ever-expanding field of gravitational wave science calls for controlled laboratory experiments on dynamic gravitation to provide insights and more precise measurements of gravity. The authors propose, model, and experimentally realize a system using two rotating bars as transmitter and a bending beam resonator as detector, finding that the near field gravitational energy transfer that creates detector amplitudes of up to 100 nm/s is 10 to the power of 25 times higher than what would be expected from gravitational waves emitted from an equivalent quadrupole gravitational wave generator.

Establishing standardized immune phenotyping of metastatic melanoma by digital pathology

The authors present a robust diagnostic algorithm based on digital pathology and image analysis that quantifies intratumoral and stromal CD8+ T-cell densities in the tumor center and invasive margin compartment in metastatic melanoma. Spatial CD8+ T-cell densities are translated into the clinically relevant immune diagnostic categories “inflamed”, “excluded”, and “desert”. Their approach also allows efficient immune phenotyping of metastatic lesions, on biopsy material or even in the absence of material from the invasive margin.
