Standard operating procedure combined with comprehensive quality control system for multiple LC-MS platforms urinary proteomics

Urinary proteomics offers sensitive, non-invasive biomarkers, but cross-platform LC-MS data comparability requires investigation. Here, the authors evaluate 20 LC-MS platforms, developing MSCohort, a QC system with 81 metrics, and a unified SOP, paving the way for precision medicine research.

Inflammasome protein scaffolds the DNA damage complex during tumor development

Shen et al. identify a noncanonical role for the inflammasome protein NLR family CARD ___domain-containing protein 4 (NLRC4) to attenuate tumor development. NLRC4 forms a scaffold to assemble the ataxia telangiectasia and Rad3-related DNA repair complex and the repair kinase checkpoint kinase-1 to promote repair of DNA breaks.

Mechanism of NAIP—NLRC4 inflammasome activation revealed by cryo-EM structure of unliganded NAIP5

The cryo-EM structure of unliganded mouse NAIP5 reveals the mechanism for NAIP—NLRC4 inflammasome activation in which ligand binding drives a roughly 20° rotation in the WHD–HD2–LRR domains, resulting in the formation of a steric clash to activate NLRC4 and generating new interactions to stabilize the NAIP5–NLRC4 complex.

Structural basis for site-specific ribose methylation by box C/D RNA protein complexes

A complex of RNA and protein known as the box C/D RNP catalyses the site-specific modification of RNAs with a 2′-O-methylation group. The structure of the full complex has now been solved, including the guide RNA and either of two substrate RNAs. This structure reveals how the guide and target RNAs are aligned, and how the methyltransferase subunit, fibrillarin, facilitates placement of the target ribose into the active site.

The NuRD complex cooperates with SALL4 to orchestrate reprogramming

Somatic reprogramming involves both transcriptional and epigenetic resetting, but we don’t yet fully understand this process. Here they show that Jdp2, Glis1, Esrrb, and Sall4 can mediate reprogramming by recruiting the NuRD complex to close chromatin, highlighting a potential role in cell fate control.
