pegRNA scaffold incorporation is a common by-product of prime editing, compromising its precision and safety. Here, the authors show that an internal abasic spacer or 2’-O-methylation within pegRNAs mitigates these by-products by terminating reverse transcriptase before reaching pegRNA scaffold.
