Cavin1 intrinsically disordered domains are essential for fuzzy electrostatic interactions and caveola formation

Caveolae are spherical nanodomains of the plasma membrane generated by assembly of caveolin and cavin proteins. Here, the authors show that fuzzy electrostatic interactions between caveolin-1 and Cavin1 proteins, combined with membrane lipid interactions, are required to generate membrane curvature and a metastable caveola coat.

Correlative cryo-ET identifies actin/tropomyosin filaments that mediate cell–substrate adhesion in cancer cells and mechanosensitivity of cell proliferation

The role of actin/tropomyosin filaments in the assembly of cell–substrate adhesions has been investigated and it is now shown by cryo-electron tomography that they are essential for adhesion assembly and also regulate mechanosensing, matrix remodelling and transformation of cells towards a cancer phenotype.

An Abl-FBP17 mechanosensing system couples local plasma membrane curvature and stress fiber remodeling during mechanoadaptation

Mechanical forces are sensed by cells and can alter plasma membrane properties, but biochemical changes underlying this are not clear. Here the authors show tension is sensed by c-Abl and FBP17, which couples changes in mechanical tension to remodelling of the plasma membrane and actin cytoskeleton.

Flotillins promote T cell receptor sorting through a fast Rab5–Rab11 endocytic recycling axis

Internalized receptors are recycled back to the cell surface, but their precise mechanisms are unclear. Here, the authors show that the flotillin membrane proteins may regulate the transfer of internalized T cell receptors into Rab5 and Rab11-positive endosomes to support its rapid recycling.

Galectin-3 drives glycosphingolipid-dependent biogenesis of clathrin-independent carriers

Many cell surface receptors are internalized by clathrin-independent endocytosis, but how clathrin-independent carriers (CLICs) are generated at the plasma membrane remained unclear. Johannes and colleagues now report that galectin-3 (Gal3) binds to glycosylated cargo proteins and glycosphingolipids. These interactions induce membrane deformation, revealing a mechanism for CLIC biogenesis.
