GP73 is a glucogenic hormone contributing to SARS-CoV-2-induced hyperglycemia

Zhong et al. show that the protein GP73 stimulates hepatic glucose production and is induced in response to infection with SARS-CoV-2 in vitro and in vivo, thus proposing a molecular mechanism underlying hyperglycemia associated with COVID-19.

Treatment of acute lung injury by targeting MG53-mediated cell membrane repair

MG53 acts as a membrane repair protein in striatal muscle. Here the authors report that MG53 is also expressed in the lung, where it exerts a similar repair function in the context of pulmonary epithelial damage, and show that administration of recombinant MG53 ameliorates lung injury in mice.

MG53-induced IRS-1 ubiquitination negatively regulates skeletal myogenesis and insulin signalling

The protein MG53 is known to inhibit myogenesis. Here, Ko et al. show that MG53 is an E3 ubiquitin ligase that mediates degradation of insulin receptor substrate 1 in skeletal muscle, thereby regulating myogenesis and insulin sensitivity in vitro and in vivo.

MG53 nucleates assembly of cell membrane repair machinery

A phosphatidylserine-binding protein, MG53, is shown to participate in membrane repair. MG53 recruits vesicles to the repair site in an oxidation dependent manner and MG53-null mice develop progressive myopathy associated with defective membrane repair.

NAADP mobilizes calcium from acidic organelles through two-pore channels

Ca²⁺ mobilization from intracellular stores represents an important cell signalling process that is regulated, in mammalian cells, by inositol-1,4,5-trisphosphate (InsP₃), cyclic ADP ribose and nicotinic acid adenine dinucleotide phosphate (NAADP). While the nature of the receptors for InsP₃ and cyclic ADP ribose are known, here the lysosomal two-pore channel, TPC2, is shown to be the molecular target of NAADP.
