Selective fluorescent probes for live-cell monitoring of sulphide

Molecular probes that can detect aqueous sulphides could help to elucidate their roles in biological signalling. Qianet al. develop two sulphide-selective fluorescent probes and demonstrate their ability to image free sulphide in living cells.

Architecture of the human G-protein-methylmalonyl-CoA mutase nanoassembly for B₁₂ delivery and repair

B₁₂-dependent human methylmalonly-CoA mutase (MMUT) requires the chaperone MMAA. The authors report the crystal structure of MMUT-MMAA, which shows a MMAA-driven conformational change in MMUT involved n B₁₂ loading and repair and helps explain the effects of disease-causing MMAA-MMUT interface mutations.

Sulfide oxidation promotes hypoxic angiogenesis and neovascularization

Hypoxia induces ·NO-dependent hydrogen sulfide (H₂S) biogenesis by inhibiting the transsulfuration pathway. H₂S oxidation promotes endothelial cell proliferation to support neovascularization in tissue injury and tumor xenograft models.

The thiostrepton A tryptophan methyltransferase TsrM catalyses a cob(II)alamin-dependent methyl transfer reaction

Cobalamin-dependent radical SAM enzymes are proposed as methyltransferases in many biosynthetic pathways. Here, the authors study a pathway involving the methylation of tryptophan, showing that methylcob(III)alamin is the most likely cofactor and propose a radical-based C-methylation mechanism.

A switch III motif relays signaling between a B₁₂ enzyme and its G-protein chaperone

Heterotrimeric G proteins contain a switch III motif that regulates enzyme function. Structural and biochemical studies now identify a similar switch III loop in a nonheterotrimeric G-protein chaperone that explains the debilitating effects of mutations linked to methylmalonic aciduria.
