RNA polymerase II depletion from the inactive X chromosome territory is not mediated by physical compartmentalization

The authors show that the apparent depletion of RNAPII from the inactive X chromosome territory is not due to a biophysical compartmentalization effect, but rather due to the loss of RNAPII bound fraction on chromatin.

C/EBPα creates elite cells for iPSC reprogramming by upregulating Klf4 and increasing the levels of Lsd1 and Brd4

Graf and colleagues find that B cells exposed to a pulse of C/EBPα and the Yamanaka factors convert into elite-type cells that rapidly and efficiently reprogram into iPSCs, in a process that involves upregulation of Lsd1, Brd4 and Klf4.

The authors analyze time-resolved changes in genome topology, gene expression, transcription-factor binding, and chromatin state during iPSC generation. They conclude that 3D genome reorganization generally precedes gene expression changes and that removal of locus-specific topological barriers explains why pluripotency genes are activated sequentially during reprogramming.
