Unraveling Alzheimer’s complexity with a distinct Aβ₄₂ fibril type and specific AV-45 binding

Two types of amyloid-β protein (1–42) (Aβ₄₂) fibrils are known to form in the insoluble fraction of diseased human brains. Now, another type of Aβ₄₂ fibril (type III) has been identified in the soluble fraction of Alzheimer’s disease brain using cryo-electron microscopy. These studies also reveal differences in ligand–fibril interactions between fibril types.

Time-course remodeling and pathology intervention of α-synuclein amyloid fibril by heparin and heparin-like oligosaccharides

Here, the authors reveal progressive conformational changes of α-synuclein fibrils upon binding with polysaccharide heparin, suggesting that biopolymers have a crucial role in remodeling the structures and pathological activities of amyloid fibrils.

Heparin induces α-synuclein to form new fibril polymorphs with attenuated neuropathology

The Cryo-EM structures reported in this work reveal how heparin incorporates into α-syn fibril formation to determine fibril polymorphs. This highlights the role of biological polymers in the conformational selection and neuropathological regulation of amyloid fibrils.

Phosphorylation and O-GlcNAcylation at the same α-synuclein site generate distinct fibril structures

Here, the authors use cryo-EM to show that phosphorylating or O-GlcNAcylating α-synuclein on serine 87 leads to the formation of two distinct fibril structures. Both structures display reduced neurotoxicity and propagation activity.

Parkinson’s disease associated mutation E46K of α-synuclein triggers the formation of a distinct fibril structure

The E46K α-synuclein mutation causes familial Parkinson’s disease. Here, the authors present the cryo-EM structure of N-terminally acetylated E46K α-synuclein fibrils and find that it is distinct from other known α-synuclein fibril structures.
