The SIAH2-NRF1 axis spatially regulates tumor microenvironment remodeling for tumor progression

Tumor cell-microenvironment interactions generate heterogeneity and promote progression. Here, Ma et al. show that the E3 ligase SIAH2 degrades NRF1 in response to hypoxia to enhance pro-tumor metabolic and environmental effects, whereas NRF1 stabilization sensitizes tumor cells to apoptosis.

Insights into solvent and surface charge effects on Volmer step kinetics on Pt (111)

Excess free charges on electrode surfaces drive changes in hydrogen electrocatalysis kinetics. Here, the authors show how redox solvent reorganization energy is insensitive to interfacial electric field strength; instead, the charged surface directly modulates proton electrochemical potential.

Nickel–molybdenum–niobium metallic glass for efficient hydrogen oxidation in hydroxide exchange membrane fuel cells

Hydroxide exchange membrane fuel cells operating in alkaline electrolyte are more cost-effective than their proton exchange membrane counterparts, but their performance is still considerably lower. Now, a Ni–Mo–Nb metallic glass is put forward as a hydrogen oxidation reaction catalyst with high activity and stability in alkaline electrolyte.

G-protein β2 subunit interacts with mitofusin 1 to regulate mitochondrial fusion

Little is known about the regulation of mitofusin proteins that are involved in the fusion of mitochondrial membranes. In this study, the authors report that a heterotrimeric G protein, Gβ-subunit 2, can interact with mitofusin 1, and is involved in the regulation of mitochondrial fusion.

Zyxin-Siah2–Lats2 axis mediates cooperation between Hippo and TGF-β signalling pathways

Hippo and TGF-β are crucial signalling pathways involved in the development of various types of tumours. Here, the authors demonstrate that TGF-β can directly regulate Hippo pathway through the stabilization of the scaffold protein Zyxin, which forms a ternary complex with Siah2 and Lats2 promoting Lats2 degradation and YAP activation.

Damaged mitochondria can be removed by mitophagy, but how this organelle is recognized by the autophagy machinery is unclear. Chen and colleagues show that FUNDC1, an integral mitochondrial outer membrane protein, interacts with light chain 3 (LC3) and is essential for hypoxia-induced mitophagy.

Wu and colleagues report that under hypoxic conditions the LATS2 kinase is targeted for degradation by the SIAH2 ubiquitin ligase, leading to inhibition of the Hippo kinase cascade and activation of YAP, which promotes tumour growth.
