NMN attenuates cuproptosis-induced DNA damage and loss of Fe-S protein POLD1. HeLa cells were cultured in DMEM with 10% copper free serum replacement and pretreated with NMN or TTM before pulsed with 1.5 μM Es/Cu for 24 h, the cells were harvest. The γH2AX Immunofluorescent staining (C). The expression of γH2AX and POLD1 protein levels were determined by western-blot (A, D) and the bands density were analyzed (B, E). Results were the mean from 3 to 5 independent experiments. Data were represented as mean ± SEM. ns, no significance; * P < 0.05; ** P < 0.01; * P < 0.001; * P < 0.0001, the DNA damage levels were assayed.
