Total liver and skeletal muscle homogenates were prepared from the vehicle-injected male WT and KO rats that were the same as used in Fig. 8. (A–D) The IRS-2 protein level in the liver (A, B) and skeletal muscle (C, D) of 8-week-old WT (n = 7) and KO (n = 7) rats with − insulin. Total liver and muscle homogenates were analyzed by immunoblotting with antibodies against IRS-2 and β-tubulin. β-tubulin was used as an internal control. (A, C) Representative immunoblots. (B, D) The immunoreactivity of IRS-2 was quantified and divided by the immunoreactivity of β-tubulin. Values are means ± SDs. *p < 0.05 vs. WT rats. Similar results were obtained using the liver and skeletal muscle homogenates from the vehicle-injected male WT and KO rats that were the same as used in Figs. 5 and 6.
