Combination immunotherapy with anti-PD-1 and anti-LAG-3 antibodies has shown anti-tumor effects across various cancers, however not all patients respond. Here the authors report that resistance to anti-LAG-3 plus anti-PD-1 therapy in head and neck cancer is mediated by the interaction of Sox9+ tumor cells with Frp1+ neutrophils, resulting in impaired anti-tumor immunity.

ITGB6 modulates resistance to anti-CD276 therapy in head and neck cancer by promoting PF4⁺ macrophage infiltration

Response rate to anti-CD276 based immunotherapy remains suboptimal in patients with cancer. Here, in a chemically-induced murine model of head and neck squamous cell carcinoma, the authors show that expression of ITGB6 in tumor cells promotes resistance to anti-CD276 therapy, associated with accumulation of PF4+ macrophages and T cell dysfunction.

CD276-dependent efferocytosis by tumor-associated macrophages promotes immune evasion in bladder cancer

Tumor associated macrophages (TAM) are playing an active role in tumor immune evasion in multiple cancer type. Here authors show that CD276 expression by TAMs may underpin this immune-suppressive role via promoting efferocytosis and suppressing MHC class II expression, which result in decreased CD4+ and CD8 + T cell infiltration.

Hybrid chalcogen bonds in prodrug nanoassemblies provides dual redox-responsivity in the tumor microenvironment

While homodimeric prodrug assemblies can improve drug loading and limit toxicity in cancer therapy, bioactivation within the target site is limited. Here, the authors introduce a hybrid chalcogen bond linker to a docetaxel dimeric prodrug nanoassembly and demonstrate its improved selfassembly, redox-responsivity and antitumor efficacy.
